The victory at the Twickenham Stoop was the Light Blues' 16th in 29 matches.
Oxford opened the scoring through pacy winger James Batstone, the two-try hero of last year's victory, but prop Matt Bray brought the Light Blues level on 14 minutes with a close-range effort.
James Hunt added the conversion and extended his side's lead to 10-4 with penalties either side of half-time.
The Dark Blues created a host of tryscoring opportunities in the second half as they sought to draw level but their handling let them down.
Cambridge , who included 11 survivors from their 2008 defeat, clinched the win with late tries through loose forward Ignacio Quintana and winger Joe Pitt-Rachid.
